The cooling bath was removed and the biphasic mixture was stirred at room temperature for 4 h. TLC analysis indicated some of the starting material still present. More thiophosgene (0.2 mL, 2.6 mmol) was added and stirring was continued for another 1 h and filtered. The organic layer was separated, and the aqueous layer was extracted with CH2Cl2. The combined organic layers were dried (Na2SO4) and concentrated to give the crude title compound as a brown solid.

The solution was filtered, and the filter cake was washed with water (100 ml x3). The filter cake was dissolved in ethyl acetate (300OmL) and dried over sodium sulfate. The solution was filtered, and the filtrate was concentrated in vacuo using a rotary evaporator. The product (204.6 g, purity: 90%, yield:78%) of 2-chloro-4-methyl- pyridin-3-ylamine was obtained as a light red solid. The material was used in next step without further purification.

When the reaction was completed for 2-3 h according to TLC analysis,the mixture was filtered, and the filtrate was diluted by CH2Cl2 (60 ml). The mixture was washed by 0.1 N HCl (30 ml), saturated Na2CO3 (30 ml) and saturated NaCl (30 ml),dried over anhydrous Na2SO4, concentrated in vacuo, and purified by PTLC to give the pure 4alpha-acyloxypodophyllotoxin derivatives 5a, b, e, f in 84-90% yields.
