A new imidazo [1,5-alpha]pyridine-based fluorescent probe, named MZC-AC, is developed for the detection of cysteine (Cys) based on an excited state intramolecular proton transfer (ESIPT) mechanism. The probe MZC-AC uses an acrylate moiety as the ESIPT blocking group as well as the recognition site. Upon the treatment with Cys, MZC-AC exhibits a dramatic fluorescence enhancement (85-fold) and a large Stokes shift (166 nm). The limit of detection obtained from fluorescent titration was as low as 0.07 mu M. MZC-AC displays highly sensitive and selective response to Cys over homocysteine (Hcy) and glutathione (GSH). Significantly, MZC-AC could be used to detect Cys in living cells.

Purpose: We investigated Nw-hydroxy L-Arginine (NOHA) predictive response in serous ovarian carcinoma based on estrogen-hormone receptor expression status; and assessed the distinctive NOHA response between estrogen-receptor-negative (ER-) tumor subtypes of ovarian and breast cancer. Materials/methods: Three-dimensional (3D) spheroids models of ER- and estrogen-receptor-positive (ER+) from breast and ovarian tumor, cultured for 9 weeks, were assayed for cellular levels of inducible nitric oxide synthase (NOS2), nitric oxide (as total nitrite) and L-Arginine, and compared to NOHA in culture medium. Statistical difference was set at p < 0.01. Results: Nine-week in vitro studies showed a progressive NOHA reduction in culture medium by at least 0.4-0.8 fold, and 0.65-0.92 fold only in the ER-breast tumor and ER-ovarian tumor 3D spheroids, respectively; with increases in cellular NOS2 and nitric-oxide levels, by at least 1.0-2.45 fold in both ER-tumor subtype 3D spheroids (p < 0.01; n = 6). Within ER-subtypes, medium NOHA decreased by >= 38.9% in ovarian cancer over breast cancer 3D-spheroids, with cellular increases in NOS2 (by >= 17.4%), and nitric oxide (by >= 18.8%). Cellular L-Arginine to medium NOHA ratio was higher, and by at least 6.5-22.5 fold in ER-breast tumor 3D-spheroids, and at least 10-70 fold in ER-ovarian tumor 3D spheroids, than in ER+ and control conditions; and was >= 48% higher in ER-ovarian cancer than in ER-breast cancer 3D-spheroids. Conclusions: The present study shows NOHA as a sensitive and selective indicator differentiating and distinguishing ER-subtypes based on the tumor grade.

Water-soluble vitamins like B3 (nicotinamide), B6 (pyridoxine), and B9 (folic acid) are of utmost importance in human health and disease, as they are involved in numerous critical metabolic reactions. Not surprisingly, deficiencies of these vitamins have been linked to various disease states. Unfortunately, not much is known about the physiological levels of B6 vitamers and vitamin B3 in an ethnically isolated group (such as an Emirati population), as well as their relationship with obesity. The aim of the present study was to quantify various B6 vitamers, as well as B3, in the plasma of obese and healthy Emirati populations and to examine their correlation with obesity. A sensitive and robust HPLC-MS/MS-based method was developed for the simultaneous quantitation of five physiologically relevant forms of vitamin B6, namely pyridoxal, pyridoxine, pyridoxamine, pyridoxamine phosphate, and pyridoxal phosphate, as well as nicotinamide, in human plasma. This method was used to quantify the concentrations of these vitamers in the plasma of 57 healthy and 57 obese Emirati volunteers. Our analysis showed that the plasma concentrations of nicotinamide, pyridoxal, and pyridoxamine phosphate in the obese Emirati population were significantly higher than those in healthy volunteers (p< 0.0001,p= 0.0006, andp= 0.002, respectively). No significant differences were observed for the plasma concentrations of pyridoxine and pyridoxal phosphate. Furthermore, the concentrations of some of these vitamers in healthy Emirati volunteers were significantly different than those published in the literature for Western populations, such as American and European volunteers. Koji amazake, prepared from rice koji, is a traditional Japanese sweet beverage. The main source of sweetness is glucose derived from rice starch following digestion by enzymes of Aspergillus oryzae during saccharification. The temperature of this process was empirically determined as 45 degrees C-60 degrees C, but no studies have systematically investigated the effect of temperature on saccharification efficiency. We addressed this in the present study by evaluating saccharification efficiency at various temperatures. We found that glucose content was the highest at 50 degrees C (100%) and was reduced at temperatures of 40 degrees C (66.4%), 60 degrees C (91.9%), and 70 degrees C (76.6%). We previously reported that 12 types of oligosaccharides are present in koji amazake; the levels of eight of these, namely nigerose, kojibiose, trehalose, isomaltose, gentiobiose, raffinose, panose, and isomaltotriose, were the highest at 50 degrees C-60 degrees C, whereas sophorose production was maximal at 70 degrees C. Based on these findings, we initially performed saccharification at 50 degrees C and then switched the temperature to 70 degrees C. The maximum amount of each saccharide including sophorose that was produced was close to the values obtained at these two temperatures. Thus, oligosaccharide composition of koji amazake is dependent on saccharification temperature. These findings provide useful information for improving the consumer appeal of koji amazake by enhancing oligosaccharide content. (C) 2018, The Society for Biotechnology, Japan. All rights reserved.

Blue emitting phosphorescent materials with high efficiency and high stability are a key requirement for the wider adoption of organic light emitting devices (OLEDs). In order to achieve triplet derived emission at the higher energy region of the electromagnetic spectrum, a series of neutral, monocyclometalated gold(iii) complexes with trifluoromethyl and trifluoromethoxy functionalized 2-phenylpyridine (ppy) derivatives, 2-anilinopyridine (apy), 2-benzoylpyridine (bpy) and 2-benzylpyridine (bepy), as the cyclometalating framework and diaryl or monoaryl alkyne as ancillary ligands have been designed and synthesized. Extensive photophysical and chemical characterization by various nuclear magnetic resonance spectroscopy techniques, elemental analysis and single crystal X-ray diffraction studies of selected compounds was carried out to confirm the structural and the chemical identity of the complexes. The emission wavelength maxima of the complexes appear in the blue/sky blue region of the electromagnetic spectrum. Detailed photophysical investigations revealed that the different emission properties of the complexes are predominantly dictated by the electronic properties of the cyclometalating ligand. The interesting photoluminescence properties along with the facile synthetic access makes this a promising concept to obtain highly suitable blue emitting gold(iii) complexes.

Here, we report a new and practical strategy for the reductive deuteration of ketones using a Mg/BrCH2CH2Br/D2O system, which affords alpha-deuterated alcohols in good yields and with almost quantitative incorporation of deuterium. Pyrido[1,2-a]indoles are known for medicinally and pharmaceutically important compounds; however, the efficient synthetic routes are scarce in the literature. We report herein a convenient and efficient route to synthesize these molecules through indium-catalyzed transannulation of pyridotriazoles with arenes. A library of compounds have been synthesized employing the method developed with various substituted pyrido[1,2-a]indole derivatives in moderate to good yields. The density functional theory study using SMDDCB-M06/6-31++G(d,p)/LANL2DZ//B3LYP/6-31G(d)/LANL2DZ method suggests that the reactions proceed via indium-carbenoid complex.

A set of 2-amino-4-aryl-4H-pyrano[3,2-h]quinoline-3-carbonitrile derivatives were prepared via a one-pot, three-component condensation reaction between the substituted hydroxyquinoline derivatives, some aryl and/or hetaryl aldehydes, and malononitrile in an ethanol/piperidine solution in a microwave irradiation environment. The structure of the prepared compounds was instituted on the foundations of their spectral data: IR, H-1 NMR, C-13 NMR, and MS. Four human cancer cell lines, MCF-7, HCT-116, HepG-2, and A549 were utilized to evaluate the antiproliferative properties of the target compounds in comparison to the positive controls, Vinblastine and Colchicine using the MTT viability assay. The cell cycle arrest behavior, detected by propidium iodide as well as the apoptosis induction, which was monitored by the flow cytometer, using the Annexin V-FITC kits, was investigated. The results illustrated that the potent cytotoxic compounds induce cell cycle arrest at the G2/M phases and trigger apoptosis in the different tested cancer cells. Finally, the structure-activity relationship (SAR) study showcases the substitution of some specific groups at the 4-, 6-, and 9-positions in the prepared 2-amino-4H-pyrano[3,2-h]quinoline derivatives, which indicates that the lipophilicity manipulates the ability of these moieties against the diverse cell lines.
