Cancer is a strong global burden with increasing numbers of diseases and ongoing anticancer drug resistance. The number of structurally novel anticancer drugs is strongly limited. They cause high costs for the social health systems. Most critical so-called multidrug resistances (MDR) are caused by transmembrane efflux pumps that transport drugs with various structures out of the cancer cells. Multidrug resistance proteins (MRPs) type 1 and 2 are found overexpressed in various kinds of cancer. There is a strong need for inhibitors of those efflux pumps. We developed novel nonsymmetrical 1,4-dihydropyridines as novel inhibitors of cancer relevant MRP types 1 and 2. The structure-dependent activities of the differently substituted derivatives were evaluated in cellular assays of respective cancer cells and are discussed. Promising candidates were identified. One candidate was demonstrated to resensitize a cisplatin resistant cancer cell line and thus to overcome the anticancer drug resistance.

Herein we report, a mononuclear, highly fluorescent zinc(ii) complex Zn(L)(N-3)(H2O) (1) that was prepared by an easy one pot method, in which the tridentate Schiff base ligand (E)-4-((2-(dimethylamino)ethylimino)methyl)-5-(hydroxymethyl)-2-methylpyridin-3-ol (HL) was generated in situ. The compound is characterized by various spectroscopic techniques, and its structure was determined by single crystal X-ray diffraction studies. DFT calculations were used to understand the electronic structures of the ligand and the complex, and TD-DFT calculations were performed to interpret the nature of the electronic transitions observed in their UV-vis spectra. In the complex, Zn(II) is found to be penta-coordinated with one azide ligand, an aqua ligand and a monoanionic tridentate N,N,O-donor ligand. In an aqueous methanol (1:9 by volume) solution, at the physiological pH (0.01 M Tris-HCl buffer, pH 7.4), compound 1 exhibits an intense greenish blue fluorescence (lambda(ex) 390 nm, lambda(em) 462 nm), whose intensity is about 17-fold stronger than that of the free ligand. Compound 1 is found to show significant DNA binding activity. The pyridoxal appended tridentate ligand can be used for the bio-imaging of Zn(II).

Noble-metal photosensitizers and water reduction co-catalysts (WRCs) still present the highest activity in homogeneous photocatalytic hydrogen production. The search for earth-abundant alternatives is usually limited by the time required to screen new catalyst combinations; however, here, we utilize newly designed and developed high-throughput photoreactors for the parallel synthesis of novel WRCs and colorimetric screening of hydrogen evolution. This unique approach allowed rapid optimization of photocatalytic water reduction using the organic photosensitizer Eosin Y and the archetypal cobaloxime WRC [Co(GL1)(2)pyCl], where GL1 is dimethylglyoxime and py is pyridine. Subsequent combinatorial synthesis generated 646 unique cobalt complexes of the type [Co(LL)(2)pyCl], where LL is a bidentate ligand, that identified promising new WRC candidates for hydrogen production. Density functional theory (DFT) calculations performed on such cobaloxime derivative complexes demonstrated that reactivity depends on hydride affinity. Alkyl-substituted glyoximes were necessary for hydrogen production and showed increased activity when paired with ligands containing strong hydrogen-bond donors.

A new unsymmetrical N2O2-tetradentate Schiff-base complex of zinc(II) was synthesized by the template reaction of pyridoxal-S-methylthiosemicarbazone and 2-hydroxy-4-methoxy-benzaldehyde as starting compounds. S-methylthiosemicarbazone (1) and zinc(II) complex [Zn(L)CH3OH] ( 2) were characterized by elemental analysis, FT-IR, UV-visible, H-1, and C-13 NMR spectra. The molecular structure of the complex (2) was determined by single crystal X-ray diffraction technique. The structure consists of a distorted square-pyramidal geometry around the central metal, Zn(II). Quantum chemical calculations were carried out using density functional theory DFT/B3LYP, 6-31G (d), and LanL2DZ basis sets for theoretical characterization of the compounds. The experimental and theoretical data were compared comprehensively. The potential energy distribution (PED) analysis was performed for the assignment of vibration frequencies. In order to support in vitro studies, molecular docking studies have been carried out so that the title compound can be an inhibitor of Epidermal Growth Factor Receptor (1 m17), and the relationship between calculated HOMO energies and docking studies has been examined. In addition, the total antioxidant capacity (as TEAC value) and free radical scavenging activity of the compounds were determined by Cupric Reducing Antioxidant Capacity (CUPRAC) and 1,1-diphenyl-2-picryl hydrazyl (DPPH) methods, respectively. Transition-metal-catalysed, redox-neutral dehydrosilylation of alkenes is a long-standing challenge in organic synthesis, with current methods suffering from low selectivity and narrow scope. In this study, we report a general and simple method for the manganese-catalysed dehydrosilylation and hydrosilylation of alkenes, with Mn-2(CO)(10) as a catalyst precursor, by using a ligand-tuned metalloradical reactivity strategy. This enables versatility and controllable selectivity with a 1:1 ratio of alkenes and silanes, and the synthetic robustness and practicality of this method are demonstrated using complex alkenes and light olefins. The selectivity of the reaction has been studied using density functional theory calculations, showing the use of an (PrPNP)-Pr-i ligand to favour dehydrosilylation, while a JackiePhos ligand favours hydrosilylation. The reaction is redox-neutral and atom-economical, exhibits a broad substrate scope and excellent functional group tolerance, and is suitable for various synthetic applications on a gram scale.

Mixed ligand nickel(II) and ruthenium(II) complexes were synthesized from pyridoxal-N-allyl-thiosemicarbazone hydrochloride and triphenylphosphine. The structures of the complexes have been characterized by elemental analysis, IR, H-1 and P-31 NMR, conductivity, magnetic moment measurements and single-crystal X-ray diffraction technique. Based on X-ray crystallographic studies, a square-planar structure has been proposed for the Ni(II) complex, in which the thiosemicarbazone ligand acts as dianionic tridentate ONS ligand. In the case of the Ru(II) complex, the thiosemicarbazone is coordinated to metal atom as a monoanionic bidentate NS donor ligand in an octahedral geometry. Antioxidant activities of the ligand and its metal complexes were calculated as their trolox equivalent antioxidant capacities (TEAC) by CUPRAC method and DPPH assay. Both the ligand and its metal complexes were found to be antioxidant and are much more antioxidant at least 2.1 times than trolox, even, ligand is 3.5 times greater than that of trolox according to CUPRAC. A linear correlation (correlation coefficient R-2 = 0.9997) appeared between the obtained TEAC values by the two antioxidant assays.
