Iridium-catalyzed enantioselective transfer hydrogenation of ketones with formic acid was developed using a prolinol-phosphine chiral ligand. Cooperative action of the iridium atom and the ligand through alcohol-alkoxide interconversion is crucial to facilitate the transfer hydrogenation. Various ketones including alkyl aryl ketones, ketoesters, and an aryl heteroaryl ketone were competent substrates. An attractive feature of this catalysis is efficient discrimination between the alkyl and aryl substituents of the ketones, promoting hydrogenation with the identical sense of enantioselection regardless of steric demand of the alkyl substituent and thus resulting in a rare case of highly enantioselective transfer hydrogenation of tert-alkyl aryl ketones. Quantum chemical calculations revealed that the sp(3)-C-H/pi interaction between an sp(3)-C-H bond of the prolinol-phosphine ligand and the aryl substituent of the ketone is crucial for the enantioselection in combination with O-H center dot center dot center dot O/sp(3)-C-H center dot center dot center dot O two-point hydrogen-bonding between the chiral ligand and carbonyl group.

The interaction between pyridoxal hydrochloride (HQ) and apoCopC was investigated using Fourier transform infrared spectroscopy (FTIR), isothermal titration calorimetry (ITC), circular dichroism (CD), fluorescence spectroscopy, three-dimensional (3D) fluorescence spectroscopy, fluorescence lifetime, TNS fluorescence and docking methods. FTIR, CD, TNS fluorescence and fluorescence lifetime experiments suggested that the apoCopC conformation was altered by HQ with an increase in the random coil content and a reduction in the beta-sheet content. In addition, the data from fluorescence spectroscopy, 3D fluorescence spectroscopy and molecular docking revealed that the binding site of HQ was located in the hydrophobic area of apoCopC, and a redshift of the HQ fluorescence spectra was observed. Furthermore, ITC and fluorescence quenching data manifested that the binding ratio of HQ and apoCopC was 1:1, and the forming constant was calculated to be (7.06 +/- 0.21) x 10(5) M-1. The thermodynamic parameters Delta H and Delta S suggested that the formation of a CopC-HQcomplex depended on the hydrophobic force. Furthermore, the average binding distance between tryptophan in apoCopC and HQ was determined by means of Forster non-radioactive resonance energy transfer and molecular docking. The results agreed well with each other. As a redox switch in the modulation of copper, the interaction of apoCopC with small molecules will affect the action of the redox switch. Microwave-assisted approach was utilized as green approach to access a series of 2-pro pylquinoline-4-carbohydrazide hydrazone derivatives 10a-j of aromatic and heteroaromatic aldehydes in highly encouraging yields. It involved four steps reaction which was initiated with ring opening reaction of isatin in a basified environment and subsequent cross-coupling with pentan-2-one to produce compound 7. Esterification of 7 in acid medium led to the formation of compound 8 which was reacted with hydrazine hydrate to access 9 which upon microwave-assisted condensed with aromatic and heteroaromatic aldehydes furnished the targeted compounds 10a-j. The structures of 10aj were confirmed by physico-chemical, elemental analyses and spectroscopic characterization which include UV, FT-IR, H-1 and C-13 NMR as well as DEPT-135. The targeted compounds 10a-j, alongside with gentamicin clinical standard, were investigated for their antibacterial efficacies using agar diffusion method. 2-Propyl-N’-(pyridine-3-ylmethylene) quinoline-4-carbohydrazide 10j emerged as the best antibacterial hydrazide-hydrazone with lowest MIC value of 0.39 +/- 0.02 – 1.56 +/- 0.02 mu g/ mL across all the organisms screened. The heterogeneous copper(II)-catalyzed oxidative cyclization of 2-pyridine ketone hydrazones was achieved in ethyl acetate at room temperature in the presence of an MCM-41-anchored bidentate 2-aminoethylamino copper(II) catalyst [MCM-41-2N-Cu(OAc) (2) ], in the presence of air as the oxidant, yielding a wide variety of [1,2,3]triazolo[1,5- a ]pyridines in mostly good to high yields. The present method was also applied to the direct one-pot synthesis of [1,2,3]triazolo[1,5- a ]pyridines from 2-acylpyridine derivatives and hydrazine monohydrate. Importantly, this supported copper(II) catalyst could be conveniently obtained via a simple procedure from easily available and inexpensive reagents, recovered by filtration of the reaction mixture, and reused at least seven times without a significant loss of catalytic activity.

A new imidazo [1,5-alpha]pyridine-based fluorescent probe, named MZC-AC, is developed for the detection of cysteine (Cys) based on an excited state intramolecular proton transfer (ESIPT) mechanism. The probe MZC-AC uses an acrylate moiety as the ESIPT blocking group as well as the recognition site. Upon the treatment with Cys, MZC-AC exhibits a dramatic fluorescence enhancement (85-fold) and a large Stokes shift (166 nm). The limit of detection obtained from fluorescent titration was as low as 0.07 mu M. MZC-AC displays highly sensitive and selective response to Cys over homocysteine (Hcy) and glutathione (GSH). Significantly, MZC-AC could be used to detect Cys in living cells.

The site-specific incorporation of deuterium (D) into small molecules is frequently used to access isotopically labeled compounds with broad utility in many research areas, such as drug development, mechanistic studies, and NMR analyses. Nevertheless, the deuteration of a stereocenter in an enantioselective manner, which could slow the metabolism and improve the bioavailability of bioactive molecules, remains challenging owing to the lack of established catalytic methods. Here, we report an asymmetric alpha-deuteration strategy for azaarenes with inexpensive D2O as the deuterium source. A cooperative visible light-driven photoredox and chiral Bronsted acid-catalyzed system using a Hantzsch ester as the terminal reductant has been developed, which enables racemic alpha-chloro-azaarenes and prochiral azaarene-substituted ketones to experience a single-electron reduction-enantioselective deuteration process. The transition metal-free method provides important chiral alpha-deuterated azaarenes in satisfactory yields with good to excellent enantioselectivities (up to 99% ee) and substantial deuterium incorporation.

Three hydroxymethyl bridged Cu(ii) complexes of a pyridoxal Schiff base ligand 4-((E)-(2-(pyridin-2-yl)ethylimino)methyl)-5-(hydroxymethyl)-2-methylpyridin-3-ol (LH) have been synthesized and characterized on the basis of spectroscopic, elctrochemical and structural properties. The X-ray crystal structures of the complexes reveal dual denticity of the ligand, bidenticity in the absence of a co-ligand as in complex1, and tridenticity in the presence of a co-ligand such as SCN-/N(CN)(2)(-)as in complexes2and3. The complexes, though binuclear in the solid state, exist as a monomeric unit in solution due to the exceptionally long axial Cu-O-hydroxymethyl(2.4-2.5 angstrom) bond. All three complexes show efficient catalytic activities towards the aerial oxidation of 3,5-ditertiarybutylcatechol (DTBCH2) withk(cat)values of 5.38 x 10(4)h(-1), 1.18 x 10(5)h(-1)and 1.06 x 10(5)h(-1)in methanol. Complexes1and2also act as a selective sulphide ion sensor withK(b)values of 6.6 x 10(3)M(-1)and 8.1 x 10(3)M(-1), respectively, while their respective L.O.D. values are 3.4 mu M and 3.2 mu M.
