The human luteinizing hormone receptor (hLH-R) is a member of the glycoprotein hormone family of G-protein-coupled receptors (GPCRs), activated by luteinizing hormone (hLH) and essentially involved in the regulation of sex hormone production. Thus, hLH-R represents a valid target for the treatment of sex hormone dependent cancers and diseases (polycystic ovary syndrome, uterine fibroids, endometriosis) as well as contraception. Screening of the Bayer compound library led to the discovery of tetrahydrothienopyridine derivatives as novel, small molecule (SMOL) hLH-R inhibitors and to the development of BAY-298, the first nanomolar hLH-R antagonist reducing sex hormone levels in vivo. Further optimization of physicochemical, pharmacokinetic, and safety parameters led to the identification of BAY-899 with an improved in vitro profile and proven efficacy in vivo. BAY-298 and BAY-899 serve as valuable tool compounds to study hLH-R signaling in vitro and to interfere with the production of sex hormones in vivo.

2-Pyridyl ketones widely appear in bioactive molecules, natural products, and are employed as precursors of chiral 2-pyridine alky/aryl alcohols or 2-aminoalkyl pyridine ligands for asymmetric catalysis. Herein, a practical method for the rapid synthesis of 2-pyridyl ketone library in continuous flow is reported, in which the 2-lithiopyridine formed by Br/Li exchange reacts with commercially available esters to obtain 2-pyridyl ketones in a good yield at short reaction time. This protocol functions broadly on a variety of esters and has been applied to the synthesis of TGF-beta type 1 receptor inhibitor LY580276 intermediate in an environmentally friendly method. It is rapid, reliable, and cost-efficient to afford diverse kinds of 2-pyridyl ketones in the compound library.

Cobalt complexes containing a salen-type pyridoxal ligand with pyridine were synthesized as a new Co (III) catalytic system for the cycloaddition of carbon dioxide. Two cobalt(III) complexes possessing a salen-type pyridoxyl ligand were synthesized by the reaction of pyridoxal ligands (pyr(2)en = (N,N’-bis (pyridoxylideneiminato)ethylene) and pyr(2)cy = (N,N’-bis(pyridoxylideneiminato)cyclohexane)) and Co (OAc)(2) and characterized by various analytical methods, including infrared spectroscopy and high-resolution mass analysis. Single-crystal X-ray crystallography analysis confirmed that the cobalt pyr(2)en complex had a distorted octahedral structure: the tetradentate Schiff base ligand binds the cobalt metal in one plane, and the metal center adopts an octahedral geometry by the additional coordination of acetate and dimethyl sulfoxide. The synthesized complexes were used as catalysts in the cycloaddition of carbon dioxide (CO2) to propylene oxide. Transition-metal-catalysed, redox-neutral dehydrosilylation of alkenes is a long-standing challenge in organic synthesis, with current methods suffering from low selectivity and narrow scope. In this study, we report a general and simple method for the manganese-catalysed dehydrosilylation and hydrosilylation of alkenes, with Mn-2(CO)(10) as a catalyst precursor, by using a ligand-tuned metalloradical reactivity strategy. This enables versatility and controllable selectivity with a 1:1 ratio of alkenes and silanes, and the synthetic robustness and practicality of this method are demonstrated using complex alkenes and light olefins. The selectivity of the reaction has been studied using density functional theory calculations, showing the use of an (PrPNP)-Pr-i ligand to favour dehydrosilylation, while a JackiePhos ligand favours hydrosilylation. The reaction is redox-neutral and atom-economical, exhibits a broad substrate scope and excellent functional group tolerance, and is suitable for various synthetic applications on a gram scale.

A novel NNO pincer type ligand (E)-N’-(phenyl(pyridin-2-yl)methylene)isonicotinohydrazide, PPMINH, was synthesized by condensation of 2-benzoyl pyridine and isonicotinylhydrazide. The ligand PPMINH on reaction with metal ions Fe(III), Co(II) and Ni(II) yielded six metal derivatives. Different analysis tools like; Elemental analyses, ESI-MS, FT-IR, molar conductivity, UV-spectra, magnetic susceptibility, NMR and EPR measurement were used. Theoretical FT-IR, H-1 NMR and C-13 NMR of ligand and complex 6 exhibits quite a good correlation coefficient. EPR spectrum of complex 6 at LNT suggests octahedral geometry around Ni(II). The ligand and complexes were screened for antibacterial activity. DFT has been carried out to optimize the geometry of the ligand and complexes. The properties most relevant to their potential action as corrosion inhibitors has been calculated all parameters of DFT from the inhibitor molecule to the metal atom. The corrosion inhibition effect of compounds on mild steel in an acidic medium was studied for 24, 48, 72 h using weight loss measurement. (C) 2021 Elsevier B.V. All rights reserved.

Fermented soy sauce is used worldwide to enhance the flavour of many dishes. Many types of soy sauce are on the market, and their differences are mostly related to the country of origin, the production process applied and the ratio of ingredients used. Consequently, several aromas, tastes, colours, and textures are obtained. Nowadays, soy sauce can also be produced without microorganisms making the process shorter and cheaper. However, flavour may be lost. We have carried out a comprehensive metabolomics analysis of volatile compounds using stir bar sorptive extraction (SBSE)-GC-MS to relate differences in volatile content to production history and origin. The results revealed major differences between fermented and non-fermented soy sauces, and a list of volatile compounds is reported as being characteristic of each type. This study was able to relate volatiles to the production process using SBSE-GC-MS and to aroma characteristics using GC-O-MS.

A chemical sensor composed of pyridoxal hydrochloride schiff base based on diarylethene (1O) was synthesized. Its photochemical properties and selectivity to ions were further studied. The chemosensor could detect cyanide effectively and is almost undisturbed by other ions. When titrating CN-, the reaction aroused a distinct change in the absorption spectrum with the color change from transparent to yellow, and the fluorescence intensity centered at 562 nm was increased 68 folds. It also exhibited a good fluorescence sensing of Mg(2+ )with high selectivity and sensitivity. Upon addition of Mg2+, its emission intensity enhanced 110 folds, with the color change from dark to bright blue. Its good spectral response could be applied to molecular logic circuit. Moreover, the chemosensor could be made into test paper strips for the qualitative and quantitative detection of CN- and Mg2+.
