A new strategy to 2-aminochromanes through catalyst-free cascade reaction of 3-trifluoroacetyl-4H-chromenes and 4H-chromene-3-carbaldehydes with cyclic secondary amines is presented. The reaction proceeds through subsequent 1,4- and 1,2-additions of amine, bimolecular elimination of trifluoroacetamide or formamide, and 6-exotrig cyclization. The latter stage is a very rare example of addition of phenols to enamines. The obtained semicyclic N,O-acetals were applied as useful precursors for the synthesis of other chromanes.

Candida albicans biofilm represents a major clinical problem due to its intrinsic tolerance to anti-fungal compounds and it has been highly related to infections in catheterized patients. Few compounds are described as able to inhibit biofilm formation or to interfere with preformed biofilm of C. albicans. Here we report the in vitro evaluation of anti-biofilm activity on C albicans ATCC 10231 of a series of new and already known amine and amide indole derivatives. Among the studied compounds, fifteen resulted active on C albicans ATCC 10231 biofilm, with BMIC50 <= 16 mu g/mL. Three of them (7, 23 and 33) showed a selectivity towards mature biofilm and the most active of them was the compound 23 (BMIC50 = 4 mu g/mL). On the other hands, two different compounds (21 and 22) were selective towards biofilm formation with BMIC50 values of 8 mu g/mL Otherwise, compounds 16 and 17 resulted active on biofilm formation, with BMIC50 of 8 mu g/mL and 2 mu g/mL respectively, and on mature biofilm with BMIC50 of 2 mu g/mL. These two last compounds also showed an interesting activity towards the planktonic cells of C albicans. A selection of the more active compounds was also evaluated on different C albicans strains (PMC1042, PMC1083 and ATCC 10261), showing a comparable or higher anti-biofilm activity, especially on mature biofilm. In vivo toxicity studies using the Galleria mellonella larvae, were finally carried out on more active indole derivatives, showing that they are poorly toxic even at the highest concentrations tested (500-1000 mu g/mL). A naphthalimide-pyridoxal conjugate 1 has been designed and synthesized. Compound 1 forms a stable greenish yellow colored gel in DMSO:H2O (8:1 v/v). Rheological study reveals that the gel is mechanically strong (G> G) over a wide range of applied strains. The morphology of the gel as determined by FESEM shows a highly cross-linked fibrous network. The gel is anion-responsive and is selectively transformed into a sol with a color change from greenish yellow to deep blue only in the presence of F- among other anions. In CH3CN, compound 1 was also sensitive to basic anions such as F- and AcO- ions. In solution, F- was differentiated from AcO- through a color change. While the yellow colored solution of 1 in acetonitrile was changed into deep blue in the presence of F-, AcO- ions gave a faint blue coloration. A similar colorimetric differentiation of F- from AcO- has been possible in CH3CN by a reusable Schiff base-linked Merrifield resin 1a or 1b.

The interaction between pyridoxal hydrochloride (HQ) and apoCopC was investigated using Fourier transform infrared spectroscopy (FTIR), isothermal titration calorimetry (ITC), circular dichroism (CD), fluorescence spectroscopy, three-dimensional (3D) fluorescence spectroscopy, fluorescence lifetime, TNS fluorescence and docking methods. FTIR, CD, TNS fluorescence and fluorescence lifetime experiments suggested that the apoCopC conformation was altered by HQ with an increase in the random coil content and a reduction in the beta-sheet content. In addition, the data from fluorescence spectroscopy, 3D fluorescence spectroscopy and molecular docking revealed that the binding site of HQ was located in the hydrophobic area of apoCopC, and a redshift of the HQ fluorescence spectra was observed. Furthermore, ITC and fluorescence quenching data manifested that the binding ratio of HQ and apoCopC was 1:1, and the forming constant was calculated to be (7.06 +/- 0.21) x 10(5) M-1. The thermodynamic parameters Delta H and Delta S suggested that the formation of a CopC-HQcomplex depended on the hydrophobic force. Furthermore, the average binding distance between tryptophan in apoCopC and HQ was determined by means of Forster non-radioactive resonance energy transfer and molecular docking. The results agreed well with each other. As a redox switch in the modulation of copper, the interaction of apoCopC with small molecules will affect the action of the redox switch. Five cobalt complexes synthesized from two aroylhydrazones were characterized by elemental analyses, thermogravimetric analysis, molar conductivity, magnetic susceptibility measurements, IR and electronic spectra. Single crystal X-ray structure of one of the complex is also reported and it got crystallized in triclinic space group P (1) over bar and the crystal structure shows a distorted octahedral geometry around the metal center. Spectral data reveal that both the aroylhydrazones are tridentate and coordinate through the azomethine nitrogen, hydrazonic oxygen, and pyridyl nitrogen. Magnetic susceptibility measurements confirm the paramagnetic nature of the Co(II) complexes and one of the complex was found to be diamagnetic in nature. Additionally, HF/6-311G(d,p)/LANL2DZ calculations were performed to predict the possible intramolecular interactions contributing to the lowering of the stabilization energy. Accordingly, pi -> pi* transitions were found to be responsible for the stabilization energy for the ligands and their cobalt complexes. To describe and discuss the chemical reactivity and stability of synthesized complexes, quantum chemical parameters like frontier orbital energies, hardness, softness, energy gap, electronegativity, chemical potential, electrophilicity, polarizability and dipole moment were calculated. Also, the main electronic structure principles such as maximum hardness, minimum polarizability, and minimum electrophilicity principles were considered to evaluate the stability of the complexes. In this paper, a series of thiosemicarbazone derivatives containing different aromatic heterocyclic groups were synthesized and the tridentate donor system of the lead compound was optimized. Most of the target compounds showed improved antiproliferative activity against MGC803 cells. SAR studies revealed that compound 5d displayed significant advantages in inhibition effect with an IC50 value of 0.031 mu M, and better selectivity between cancer and normal cells than 3-AP and DpC (about 15- and 5-fold improved respectively). Besides, compound 5d showed selective antiproliferative activity in not only other cancer cells but also different gastric cancer cell lines. In-depth mechanism studies showed that compound 5d could induce mitochondria-related apoptosis which might be related to the elevation of intracellular ROS level, and cause cell cycle arrest at S phase. Moreover, 5d could evidently suppress the cell migration and invasion by blocking the EMT (epithelial-mesenchymal transition) process. Consequently, our studies provided a lead optimization strategy of thiosemicarbazone derivatives which would contribute to discover high-efficiency and low-toxicity agents for the treatment of gastric cancer. A new unsymmetrical N2O2-tetradentate Schiff-base complex of zinc(II) was synthesized by the template reaction of pyridoxal-S-methylthiosemicarbazone and 2-hydroxy-4-methoxy-benzaldehyde as starting compounds. S-methylthiosemicarbazone (1) and zinc(II) complex [Zn(L)CH3OH] ( 2) were characterized by elemental analysis, FT-IR, UV-visible, H-1, and C-13 NMR spectra. The molecular structure of the complex (2) was determined by single crystal X-ray diffraction technique. The structure consists of a distorted square-pyramidal geometry around the central metal, Zn(II). Quantum chemical calculations were carried out using density functional theory DFT/B3LYP, 6-31G (d), and LanL2DZ basis sets for theoretical characterization of the compounds. The experimental and theoretical data were compared comprehensively. The potential energy distribution (PED) analysis was performed for the assignment of vibration frequencies. In order to support in vitro studies, molecular docking studies have been carried out so that the title compound can be an inhibitor of Epidermal Growth Factor Receptor (1 m17), and the relationship between calculated HOMO energies and docking studies has been examined. In addition, the total antioxidant capacity (as TEAC value) and free radical scavenging activity of the compounds were determined by Cupric Reducing Antioxidant Capacity (CUPRAC) and 1,1-diphenyl-2-picryl hydrazyl (DPPH) methods, respectively. A method for the generation of 5-isoxazolidi-nones with exocyclic phosphonium ylide functionalities via [2+3] cycloaddition of Ph3PCCO and aldonitrones has been developed and applied in the synthesis of 4-alkylidene-5-isoxazolidinones via Wittig olefination. Background De novo lipogenesis (DNL) is the primary metabolic pathway synthesizing fatty acids from carbohydrates, protein, or alcohol. Our aim was to examine associations of in vivo levels of selected fatty acids (16:0, 16:1n7, 18:0, 18:1n9) in DNL with incidence of type 2 diabetes (T2D). Methods and findings Seventeen cohorts from 12 countries (7 from Europe, 7 from the United States, 1 from Australia, 1 from Taiwan; baseline years = 1970-1973 to 2006-2010) conducted harmonized individual-level analyses of associations of DNL-related fatty acids with incident T2D. In total, we evaluated 65,225 participants (mean ages = 52.3-75.5 years; % women = 20.4%62.3% in 12 cohorts recruiting both sexes) and 15,383 incident cases of T2D over the 9-year follow-up on average. Cohort-specific association of each of 16:0, 16:1n7, 18:0, and 18:1n9 with incident T2D was estimated, adjusted for demographic factors, socioeconomic characteristics, alcohol, smoking, physical activity, dyslipidemia, hypertension, menopausal status, and adiposity. Cohort-specific associations were meta-analyzed with an inverse-varianceweighted approach. Each of the 4 fatty acids positively related to incident T2D. Relative risks (RRs) per cohort-specific range between midpoints of the top and bottom quintiles of fatty acid concentrations were 1.53 (1.41-1.66; p< 0.001) for 16:0, 1.40 (1.33-1.48; p< 0.001) for 16:1n-7, 1.14 (1.05-1.22; p = 0.001) for 18:0, and 1.16 (1.07-1.25; p< 0.001) for 18:1n9. Heterogeneity was seen across cohorts (I-2 = 51.1%-73.1% for each fatty acid) but not explained by lipid fractions and global geographical regions. Further adjusted for triglycerides (and 16:0 when appropriate) to evaluate associations independent of overall DNL, the associations remained significant for 16:0, 16:1n7, and 18:0 but were attenuated for 18:1n9 (RR = 1.03, 95% confidence interval (CI) = 0.94-1.13). These findings had limitations in potential reverse causation and residual confounding by imprecisely measured or unmeasured factors. Conclusions Concentrations of fatty acids in the DNL were positively associated with T2D incidence. Our findings support further work to investigate a possible role of DNL and individual fatty acids in the development of T2D. Category: pyridine-derivatives. A new three-component strategy from aldehyde, p-toluenesulfonyl hydrazide and potassium thiocyanate for the synthesis of 2-aminothiadiazoles promoted by I-2 under metal-free conditions has been described. Potassium thiocyanate was used as an odorless and low-toxicity sulfur source. A wide range of aromatic aldehydes can smoothly undergo the three-component cyclization reaction under the optimized conditions to give the corresponding products in moderate to good yields.
