Tripodal ligands with main group bridghead units are well established in coordination chem. and single-site organometallic catalysis. Although a large number of tris(2-pyridyl) main group ligands [E(2-py)3] (E = main group element, 2-py = 2-pyridyl) spanning across the whole p-block are now synthetically acessible, only limited work was done on the coordination chem. on the tris(2-pyridyl) Group 15 ligands for the heavier elements (As, Sb). In the current study the authors study the coordination chem. of the ligand family E(6-Me-2-py)3 (E = As, Sb) and of the As(V) ligand O=As(6-Me-2-py)3. The air- and moisture-stability of all of these main group ligands makes them especially attractive in future catalytic applications.

Plajer, Alex J.;Percastegui, Edmundo G.;Santella, Marco;Rizzuto, Felix J.;Gan, Quan;Laursen, Bo W.;Nitschke, Jonathan R. research published 《 Fluorometric Recognition of Nucleotides within a Water-Soluble Tetrahedral Capsule》, the research content is summarized as follows. The design of aqueous probes and binders for complex, biol. relevant anions presents a key challenge in supramol. chem. Herein, a tetrahedral assembly with cationic faces and corners is reported that is capable of discriminating between anionic and neutral guests in water. Electrostatic repulsion between subcomponents can be overcome by the addition of an anionic template, or generating a robust covalent framework by incorporating tris(2-aminoethyl)amine (TREN). The resultant TREN-capped, water-soluble, fluorescent cage binds mono- and poly-phosphoric esters, including nucleotides. Its covalent skeleton renders it stable at micromolar concentrations in water, enabling the fluorometric detection of biol. relevant guests in an aqueous environment. Selective supramol. encapsulants, such as 1, could enable new sensing applications, such as recognition of toxins and drugs, under biol. conditions.

Novel extended side chain nitroimidazooxazine analogs featuring diverse linker groups between two aryl rings, e.g., I, were studied as a potential strategy to improve solubility and oral activity against chronic infection by Mycobacterium tuberculosis. Both lipophilic and highly polar functionalities (e.g., carboxamide, alkylamine, piperazine, piperidine, but not sulfonamide) were well tolerated in vitro, and the hydrophilic linkers provided some solubility improvements, particularly in combination with pyridine rings. Most of the 18 compounds further assessed showed high microsomal stabilities, although in the acute infection mouse model, just one stilbene (6-fold) and two pyridine-containing acetylene derivatives (5-fold and >933-fold) gave in vivo efficacies notably superior to the clin. stage compound pretomanid (PA-824). The most efficacious analog, I, also displayed outstanding in vivo activity in the stringent chronic model (up to 24-fold better than the drug delamanid and 4-fold greater than our previous best phenylpyridine candidate), with favorable pharmacokinetics, including good oral bioavailability in the rat.

Pai, Sandesh;Schott, Marco;Niklaus, Lukas;Posset, Uwe;Kurth, Dirk G. research published 《 A study of the effect of pyridine linkers on the viscosity and electrochromic properties of metallo-supramolecular coordination polymers》, the research content is summarized as follows. The authors present the optical, electrochem., and electrochromic properties of Fe(II)-, Co(II)- and Ru(II)-based metallo-supramol. polymers (MEPEs) self-assembled from rigid, π-conjugated bis-terpyridines with different numbers of pyridine linkers. By exciting the metal-to-ligand charge transfer (MLCT) transition, Ru-MEPEs show a weak luminescence in the NIR region (λlum = 765 nm) at room temperature The viscosity of Co-MEPEs in water, ethanol and 75% acetic acid is significantly higher in comparison to the analogous Fe-MEPEs. The SEM cross-section images of the highly transparent dip-coated films show smooth and homogenous surfaces with film thicknesses of <200 nm for Fe-, Co- and Ru-MEPEs. The cyclic voltammograms of the Fe- and Ru-MEPEs show a reversible redox couple corresponding to the Fe(II)/Fe(III) and Ru(II)/Ru(III) states, resp. Co-MEPEs display two metal centered Co(II)/Co(I) and Co(II)/Co(III) reversible redox waves at -1.12 and -0.10 V vs. Fc/Fc+, resp. Fe-MEPE films show a promising high cycle stability over 1000 switching cycles. The coloration efficiencies were also determined for Fe-MEPEs (720 mC cm-2 at 594 nm). The study of the electrochromic properties reveals that the introduction of pyridine linkers enhances the switching stability and reversibility. Co-MEPEs exhibit a broad absorption band covering the visible and NIR region at a neg. potential (-1.4 V vs. Fc/Fc+) due to the metal-to-ligand charge transfer from Co(I) to the acceptor terpyridine ligands.

Ozenil, Marius;Pacher, Katharina;Balber, Theresa;Vraka, Chrysoula;Roller, Alexander;Holzer, Wolfgang;Spreitzer, Helmut;Mitterhauser, Markus;Wadsak, Wolfgang;Hacker, Marcus;Pichler, Verena research published 《 Enhanced arecoline derivatives as muscarinic acetylcholine receptor M1 ligands for potential application as PET radiotracers》, the research content is summarized as follows. Supported by their involvement in many neurodegenerative disorders, muscarinic acetylcholine receptors (mAChRs) are an interesting target for PET imaging. Nevertheless, no radiotracer is established in clin. routine. Within this work we aim to develop novel PET tracers based on the structure of arecoline. Fifteen novel arecoline derivatives were synthesized, characterized and tested for their affinity to the mAChRs M1-M5 and the conceivable off-target acetylcholinesterase. Five arecoline derivatives and arecoline were labeled with carbon-11 in good yields. Arecaidine diphenylmethyl ester (3b), arecaidine bis(4-fluorophenyl)methyl ester (3c) and arecaidine (4-bromophenyl)(4-fluorophenyl)methyl ester showed a tremendous gain in mAChR affinity compared to arecoline and a pronounced subtype selectivity for M1. Metabolic stability and serum protein binding of [11C]3b and [11C]3c were in line with properties of established brain tracers. Nonspecific binding of [11C]3c was prevalent in kinetic and endpoint experiment on living cells as well as in autoradiog. on native mouse brain sections, which motivates us to decrease the lipophilicity of this substance class prior to in vivo experiments

Jean-Luc;Bishai, William R. research published 《 Design, synthesis, and biological evaluation of novel imidazo[1,2-a]pyridinecarboxamides as potent anti-tuberculosis agents》, the research content is summarized as follows. Tuberculosis (TB) is a highly infectious disease that has been plaguing the human race for centuries. The emergence of multidrug-resistant strains of TB has been detrimental to the fight against tuberculosis with very few safe therapeutic options available. As part of an ongoing effort to identify potent anti-tuberculosis agents, we synthesized and screened a series of novel imidazo[1,2-a]pyridinecarboxamide derivatives for their anti-tuberculosis properties. These compounds were designed based on reported anti-tuberculosis properties of the indolecarboxamides (I2Cs) and imidazo[1,2-a]pyridinecarboxamides (IPAs). In this series, we identified compounds 15(I) and 16(II) with excellent anti-TB activity against H37Rv strain of tuberculosis (MIC = 0.10-0.19 μM); these compounds were further screened against selected clin. isolates of Mtb. Compounds 15 and 16 showed excellent activities against multidrug-resistant (MDR) and extensively drug-resistant (XDR) strains of TB (MIC range: 0.05-1.5 μM) with excellent selectivity indexes. In addition, preliminary ADME studies on compound 16 showed favorable pharmacokinetic properties.
