The purpose of the research is to develop a reverse-phase high performance liquid chromatog.(RP-HPLC) method to determine pheniramine maleate concentration in the pheniramine maleate-naphazoline hydrochloride eye drops. The HPLC separation was carried out on an Agilent C18(150 mm*4.6 mm, 5μm) column, using a UV detector with 262 nm wavelength. The mobile phase was 0.1 mol·L-1 ammonium acetate buffer(0.5% triethylamine, adjusted to pH 4.0 with acetic acid)-methanol solution=97:3(V/V), with velocity of 1 mL·min-1 and column temperature of 20°C. The good linearity of pheniramine maleate under chromatog. was found(r>0.999, n=7). The recovery values were between 97% and 103%(n=3). The RSD values were below 2.0%(n=3). The sample was stable in 24 h. The samples reached standardized qualification. RP-HPLC is proved to be a simple, swift, accurate and reliable method and has good reproducibility.
