A novel run of fused heterocyclic derivatives containing pyridine moieties has been disclosed by allowing 2-amino-4-phenyl-6-(phenyl amino)pyridine-3,5-dicarbonitrile 1 to undergo annulation reactions with different reagents. Most of synthesized compounds have moderate to strong antitumor activity against HePG-2 and MCF-7. Moreover, MOE 2014.09 software was used to run the computational studies to support the biological activity results. The assigned structures for all the newly prepared derivatives were ascertained on the basis of elemental analyses and spectral data.

Mono- and dialkenyl-substituted 2-(1H-pyrazol-1-yl)pyridines could be selectively synthesized in moderate to good yields by tuning the reaction conditions. Further synthetic transformations were conducted to demonstrate the synthetic potential of our products.

Synthesis of New Derivatives of 5-Acetyl-4-hydroxy-2H-1,3-thiazine-2,6(3H)-dione

The condensation of the title compound with benzene-1,2-diamine in propan-2-ol, followed by addition of 5-arylfuran-2-carbaldehyde and trifluoroacetic acid afforded 5-(2-{5-[4-bromo(nitro)phenyl]furan-2-yl}-2,3-dihydro-1H-1,5-benzodiazepin-4-yl)-4-hydroxy-2H-1,3-thiazine-2,6(3H)-diones. 5-((E)-3-{5-[4-Bromo(nitro)phenyl]furan-2-yl}prop-2-enoyl)-4-hydroxy-2H-1,3-thiazine-2,6(3H)-diones were synthesized by condensation of 5-acetyl-4-hydroxy-2H-1,3-thiazine-2,6(3H)-dione with 5-[4-bromo(nitro)phenyl]furan-2-carbaldehydes in chloroform in the presence of catalytic amounts of pyridine and piperidine.

In contrast, the addition of pyridine to the Co-catalyzed system showed the opposite chemoselectivity, affording the ketone hydrosilylated product. The chemoselectivity of olefin/ketone hydrosilylation was switched by replacing the central metal of the complex with the iminobipyridine derivative ligand and by changing the simple reaction conditions (absence or presence of pyridine) using an identical Co complex.
