The reaction mixture was cooled to room temperature, and ethyl acetate and water were then added thereto. The organic layer was separated, washed with a saturated aqueous sodium chloride solution and dried over anhydrous sodium sulfate, and the solvent was distilled off under reduced pressure. The obtained residue was purified by silica gel column chromatography (gradient elution with hexane:ethyl acetate = 100:0-50:50). Water and methanol were added to the thus obtained residue, and the solid was collected by filtration to give 91 mg of methyl 2-((1-benzyl-1H-indol-5-yl)amino)-5-(trifluoromethyl)nicotinate as a yellow solid. 1H-NMR (DMSO-d6) delta: 3.93 (3H, s), 5.43 (2H, s), 6.48 (1H, d, J = 2.6 Hz), 7.18-7.34 (6H, m), 7.43 (1H, d, J = 8.6 Hz), 7.54 (1H, d, J = 3.3 Hz), 7.88 (1H, d, J = 2.0 Hz), 8.38 (1H, d, J = 2.6 Hz), 8.66 (1H, d, J = 2.0 Hz), 10.20 (1H, s). MS (ESI, m/z): 426 (M+H)+.
