The detection of Cu2+ in living plants and animals is of great importance for environment monitoring and disease diagnosis. Here, a near-IR (NIR) turn-on photoacoustic (PA) probe (denoted as LET-2) is developed for Cu2+ detection in living subjects, such as soybean sprouts and mice. Upon reaction with HNO, CuII[TD1] shows a 16.1-fold turn-on in EPR signal with a low detection limit of 1.95 μM. Moreover, low-temperature EPR spectroscopic and ESI-MS studies showed that the sensing mechanism relies on the reduction of Cu(II) by HNO. Lastly, CuII[TD1] is selective for HNO over other reactive nitrogen and oxygen species except for some reductants (Cys and Asc). This new Cu(II) coordination-containing TAM radical shows great potential for in vivo EPR HNO applications in the absence of reducing agents and provides insights into developing improved and targeted EPR HNO probes for biomedical applications. The success of the chem. reaction and biomol. recognition process that occur in a confined geometry can be monitored from the changes in elec. readout of the nanochannel. The nanofluidic sensor has the ability to sensitively and specifically detect lower concentrations (≥1 nM) of phosphoprotein (albumin and α-casein) in the surrounding environment as evidenced from the significant decrease in ion current flowing through the nanochannels. However, dephosphoproteins such as lysozyme and dephospho-α-casein even at higher concentration (>1μM) could not induce any significant change in the transmembrane ion flux. This observation indicated the sensitivity and specificity of the proposed nanofluidic sensor towards PPn proteins, and has potential for use in differentiating between phosphoproteins and dephosphoproteins.
