The release of Fe from horse spleen ferritin (I) by the chelating agents, deferrioxamine B (II), rhodotorulic acid (III), 2,3-dihydroxybenzoate (IV), 2,2′-bipyridyl (V), and pyridine-2-aldehyde 2-pyridylhydrazone (VI), was studied in vitro. I prepared by classical procedures involving thermal denaturation released its Fe less effectively than I isolated by a modified procedure that avoided this step. II and III were the most effective in releasing Fe from both preparations of I. When FMN was added, Fe release by II, III, and IV was effectively blocked, whereas both V and VI showed a marked simulation. A substantial increase in Fe release was also observed for V and VI with ascorbate; a less important increase was noted for III. EDTA exerted a marked inhibition of Fe release from I with III, IV, V, and VI. The effects of citrate and oxalate on Fe release by the chelators was small. The effect of the concentration of flavin on Fe release from I by II and V was studied. II was unable to mobilize Fe(II) from I following reduction by reduced FMN, whereas V could rapidly complex Fe(II). The results are discussed in the context of current concepts of storage Fe mobilization in the treatment of Fe overload.

We describe a study of the equilibrium binding of Zn2+ to binary mixtures of tridentate ligands terpyridine (1), 2,5-(CHNC6H5)2pyridine (2) and (2-pyridyl)NHNCH(2-pyridyl) (3) which leads to the formation of a dynamic mixture of homo ([Zn(1)2](CF3SO3)2 4, [Zn(2)2](CF3SO3)2 5, [Zn(3)2](CF3SO3)2 6) and hetero ([Zn(1)(3)](CF3SO3)2 7, [Zn(1)(2)](CF3SO3)2 8, [Zn(2)(3)](CF3SO3)2 9) coordination compounds We report the crystal structures of five such complexes (4, 5, 79) which assemble into complementary duplex compounds that further self-organize into double helical- or lamellar-type architectures in the solid state. The ligand exchange between homo-duplex complexes in solution leads to the preferential formation of the heteroduplex complexes. As might be expected these processes display a statistical distribution of homo-duplex:heteroduplex:homo-duplex complexes of 1:2:1 for the mixtures resulting from 4:6 and 5:6 ligands. However, complex 8 is preferred in a 4:5 mixture which presents a composition of 1:4:1 (amplification factor of about 33%). The X-ray structural determinations of selected single crystals resulting from the stoichiometric mixtures of homo-duplex complexes show unique heteroduplex superstructures 79 in the solid state. The present results present the solid-state structures of homo- and hetero-complexes resulting from terpyridine-type ligands 13 and Zn2+ metal ions. The heteroduplex Zn2+ complexes 79 are quant. crystallized in the solid state by statistical (7, 9) and structural (8) driven selection in solution from a binary mixture of the terpyridine-type complexes and then trapped by crystallization in the solid state.

The equilibrium and kinetics of the extraction of Ni with benzene solutions of 2-pyridinecarbaldehyde 2-(5-substituted)pyridylhydrazones were studied at 25 ± 0.1° and an ionic strength of 0.2. The extracted species was found too be NiL2 for each hydrazone, where L denotes the undissociable part of the hydrazone. Partition constants of the hydrazones and extraction constants of their Ni complexes between an aqueous and a benzene phase were determined The rate of extraction was best described by a two-term expression, both terms being 1st order in Ni ion and HL concentrations The first term has zero dependence on H+ concentration, whereas the 2nd is inversely first order with respect to H+ concentration These results support the hypothesis that the rate-determining steps involve 2 concurrent reactions of Ni with neutral and anionic forms of the hydrazones. Kinetic data and activation parameters for the first reaction were determined, but those for the second one involving the anion could not be obtained.

This is attributed to an intramol. hydrogen bond between the imino hydrogen and the nitrogen of pyridine of the latter hydrazones.

Solvent extraction of nickel(II) complexes with five heterocyclic hydrazones (containing pyridine- or/ and quinoline-nitrogen atoms as the coordinating sites) into chloroform or benzene was studied kinetically at 25°C and μ = 0.1 by means of a spectrophotometric method. The kinetics of extraction of nickel(II) ion is first order in each of the aqueous nickel(II) concentration and the hydrazone concentration in the organic phase. The observed second-order rate constant, kobs, -pH profiles suggest three rate-determining steps to be considered in the aqueous phase: Ni2+ + H2L+ â†? Ni2+ + HL â†?and Ni(OH)+ + HL â†? The rate constant for the formation of 1:1 complexes with 2-pyridylmethanone 2-pyridylhydrazone (PAPH) was consistent with those for bipyridine and terpyridine. Replacement of pyridyl group by quinolyl group affects the distribution constant (Kd) of the ligand between the two phases rather than the rate constants The influence of the structure of ligands on the rate constants is also discussed.

Three new hydrazones, in which the hydrogen at the 5-position of the pyridine ring in the hydrazine moiety of 2-pyridinecarboxaldehyde 2-pyridylhydrazone was substituted by a Me, chloro, or nitro group, have been synthesized and their chromogenic properties, reactivities with metal ions, and extractabilities of the metal complexes were studied. The acid dissociation constants (Ka) of the synthesized hydrazones were determined spectrophotometrically. These values obeyed Hammett’s law, the inductive effect of the substituents being found to reflect the pKa values. Of the synthesized hydrazones, 2-pyridinecarboxaldehyde 2-(5-nitro)pyridylhydrazone (PANPH), in which an electron-withdrawing nitro group was introduced, was most sensitive for metal ions. Its copper and nickel complexes showed molar absorptivities of the order of 105. A highly sensitive and selective extraction-spectrophotometric method for the determination of nickel with PANPH is proposed and has been successfully applied to the determination of nickel in steel samples. The method has been further sensitized by employing analog derivative spectrophotometry.

The equilibrium of nickel(II) ion with thirteen nitrogen-heterocyclic hydrazones (neutral form: HL) with and without blocking group(s) have been investigated spectrophotometrically in 28% aqueous dioxane solution at 25°C and an ionic strength of 0.2 (KCl). For a series of 2-pyridinecarbaldehyde 5-substituted-2-pyridylhydrazones, the following linear free energy relationships (LFER) were derived between the overall formation constants, β1, β1‘, and β2, of Ni(HL)2+, NiL+, and Ni(HL)22+ complexes, resp., and the dissociation constants, Ka1 and Ka2, of H3L2+ and H2L+, resp.: log β1 = 0.95 (pKa1 + pKa2)/2 + 4.60, 1/2 log β2 = 0.95 × (pKa1 + pKa2)/2 + 4.82, and log β1‘ = 1.40 (pKa1 + pKa2)/2 + 9.73. The steric effects of the 6-Me group in pyridine and the quinolyl ring in place of pyridyl group on the complex formation were evaluated on the basis of LFER. The complex stabilization effect of the pyridyl group substituted for formyl hydrogen was also evaluated for Ni(HL)22+, Ni(HL)2+, and NiL+ complexes. These results together with LFER were discussed in conjunction with the thermodn. parameters for the formation of Ni(HL)2+ complex and the ligand field parameters, Dq, for Ni(HL)2+ and Ni(HL)22+ complexes.
