This resulted in 35 g (81%) of 2-(6-bromopyridin-2-yl)propan-2- ol as yellow oil.

Evaluation of the effects of the T-type calcium channel enhancer SAK3 in a rat model of TAF1 deficiency

The TATA-box binding protein associated factor 1 (TAF1) is part of the TFIID complex that plays a key role during the initiation of transcription. Variants of TAF1 are associated with neurodevelopmental disorders. Previously, we found that CRISPR/Cas9 based editing of the TAF1 gene disrupts the morphology of the cerebral cortex and blunts the expression as well as the function of the CaV3.1 (T-type) voltage gated calcium channel. Here, we tested the efficacy of SAK3 (ethyl 8′-methyl-2′, 4-dioxo-2-(piperidin-1-yl)-2’H-spiro [cyclopentane-1, 3’imidazo [1, 2-a] pyridine]-2-ene-3-carboxylate), a T-type calcium channel enhancer, in an animal model of TAF1 intellectual disability (ID) syndrome. At post-natal day 3, rat pups were subjected to intracerebroventricular (ICV) injection of either gRNA-control or gRNA-TAF1 CRISPR/Cas9 viruses. At post-natal day 21, the rat pups were given SAK3 (0.25 mg/kg, p.o.) or vehicle for 14 days (i.e. till post-natal day 35) and then subjected to behavioral, morphological, and molecular studies. Oral administration of SAK3 (0.25 mg/kg, p.o.) significantly rescued locomotion abnormalities associated with TAF1 gene editing. SAK3 treatment prevented the loss of cortical neurons and GFAP-positive astrocytes observed after TAF1 gene editing. In addition, SAK3 protected cells from apoptosis. SAK3 also restored the Brain-derived neurotrophic factor/protein kinase B/Glycogen Synthase Kinase 3 Beta (BDNF/AKT/GSK3 beta) signaling axis in TAF1 edited animals. Finally, SAK3 normalized the levels of three GSK3 beta substrates CaV3.1, FOXP2, and CRMP2. We conclude that the T-type calcium channel enhancer SAK3 is beneficial against the deleterious effects of TAF1 gene-editing, in part, by stimulating the BDNF/AKT/GSK3 beta signaling pathway.

Phosphorescent iridium(III) complexes bearing L-alanine ligands: Synthesis, crystal structures, photophysical properties, DFT calculations, and use as chemosensors for Cu2+ ion

The solid-state structures of the developed Ir-1 and Ir-2 were authenticated by single X-ray diffraction. Ir(III) complexes Ir-1 and Ir-2 exhibit intense yellow and red emission respectively with high quantum efficiencies (Phi(em) = 0.88 for Ir-1, Phi(em) = 0.29 for Ir-2) in oxygen-free acetonitrile solution at room temperature. Theoretical calculations were carried out to provide a further study of the orbital distributions and electronic states of the two complexes. The two complexes displayed significant reversible phosphorescence quenching upon binding to Cu2+ ion and the binding stoichiometry was approximately 1:1.H-1 NMR titration experiments indicated that coordination of Cu2+ to the nitrogen atoms of the L-alanine ligands of the complexes induce the significant variations in optical signals. This work showed that new phosphorescent probes based on iridium complexes could be realized by introducing simple amino acid ancillary ligands.

Preparation and Characterization of a Novel Magnetic Nano Catalyst for Synthesis and Antibacterial Activities of Novel Furan-2(5H)-Ones Derivatives

The catalytic activity of the magnetic catalyst was investigated through the one-pot synthesis of novel 3,4,5-trisubstituted furan-2(5H)-ones derivatives from aryl aldehydes, 4-amino pyridine, and dimethyl acetylenedicarboxylate. The structure of all the new synthesized derivatives was determined by FT-IR, H-1-NMR, and C-13-NMR. The synthetic compounds exert moderate activity against both Gram-positive and Gram-negative bacteria and their effectiveness is higher against Bacillus subtilis. This green nano catalytic procedure has good reversibility and provides clean production in a short reaction time. These characters make the method practical and economical for researchers.

Neuroprotective effects of Suhexiang Wan on the in vitro and in vivo models of Parkinson’s disease

OBJECTIVE: To examine the role of KSOP1009 (a modified formulation of Suhexiang Wan essential oil) in an animal model of Parkinson’s disease (PD) induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydro- pyridine (MPTP) injection. METHODS: Cell toxicity, apoptosis, and reactive oxygen species (ROS) levels were analyzed in the human neuroblastoma cell line SH-SY5Y. After that, changes in animal behavior and tyrosine hydroxylase (TH) protein levels in the substantia nigra (SN) of MPTP-injected mice were examined. Three different doses of KSOP1009 (30, 100, and 300 mg/kg, n = 8 for each group) were administered daily for 7 d before MPTP injection and 14 d after MPTP injection, totaling 21 d. RESULTS: MPP+, the active metabolite of MPTP, decreased the viability of SH-SY5Y cells, whereas KSOP1009 alleviated MPP +-induced cytotoxicity. KSOP1009 (10 and 50 mg/mL) reduced MPP +-induced ROS generation compared with the control group. Treatment with 1 mM MPP + increased the percentage of depolarized/live cells, whereas KSOP1009 intake at a dose of 10 mg/mL decreased the percentage of these cells. The mean latency to fall in the rotarod test was reduced in mice treated with MPTP compared with the control group. However, mice receiving three different doses of KSOP1009 performed better than MPTP-treated animals. MPTP-treated mice were more hesitant and took longer to traverse the balance beam than the control animals. In contrast, KSOP1009-treated mice performed significantly better than MPTP-treated mice. Furthermore, the KSOP1009-treated groups had a significantly higher number of TH-positive neurons in the lesioned SN and significantly higher expression of TH in the striatum than the MPTP-treated group. MPTP treatment strongly induced Jun-N-terminal kinase (JNK) activation, whereas KSOP1009 suppressed MPTP-induced JNK activation. In addition, KSOP1009 intake reversed the decrease in the phosphorylation levels of cAMP-response element-binding protein in the brain of MPTP-treated mice. KSOP1009 also restored the decrease in dopaminergic neurons and dopamine levels in the brain of MPTP-treated mice. CONCLUSION: KSOP1009 protected mice against MPTP-induced toxicity by decreasing ROS formation and restoring mitochondrial function. (C) 2019 JTCM. All rights reserved.
