The combined organic layers were washed with water and brine consecutively (3 x), dried over Na2S04, filtered, concentrated and purified by CC (PE/EA = 5/1) to give compound P45 (1.4 g, 11%) as a yellow solid.

To a solution of 5-bromo-3-(trifluoromethyl)-2(1H)-pyridinone (D16) (2 g) and silver carbonate (6.84 g) in toluene (25 mL) stirred in air at room temperature was added 2-iodopropane (5.79 mL) dropwise. The reaction mixture was stirred at room temperature for 1 day. The reaction mixture was filtered and the filtrate was concentrated, the residue was purified by column chromatography to give 5-bromo-2-[(1-methylethyl)oxy]-3-(trifluoromethyl)pyridine (D17) (1.2 g).

Methyl iodide (0.12 mL, 1.88 mmol) and Cs2C03(0.942 g, 2.89 mmol) were added to a solution of 5-bromo-3-(trifluoromethyl)-1 H-pyridin-2-one (0.350 g, 1.45 mmol) in DMF (3 mL) and the resulting mixture was stirred for 18 h at rt. The solvent was evaporated under reduced pressure, water (10 ml) was added to the residue and the aqueous phase was extracted with EtOAc (2 x 30 mL). The combined organic phases were washed with brine and dried over MgS04, filtered and concentrated under reduced pressure. The material was triturated with ether then dried to afford title compound, which was used in the next step without any further purification (0.32 g, 86%).1H NMR (500 MHz, CDCI3) delta 7.78 (d, J = 2.1 Hz, 1 H), 7.64 (d, J = 2.7 Hz, 1 H), 3.59 (s, 3H).

The organic layer was washed with sat. aq. NaHC03 and brine consecutively, dried over NaS04, filtered, concentrated and purified by CC (PE/EA = 5/1) to give compound P46a (10.5 g, 88%) as a yellow liquid.
