615 mg (987 mumol, 50% purity) 5-tert.-butyl-2-ethyl 6,7-dihydro-4H-thieno[3,2-c]pyridine-2,5-dicarboxylate are dissolved in 4 ml of a mixture of TFA and DCM (v/v 1:1) and stirred for 30 minutes at RT. The reaction mixture is neutralised by the addition of TEA and evaporated down i. vac.The crude product thus obtained is dissolved in 4 ml formic acid and combined with 0.5 ml (6.7 mmol) formalin solution (37% in water). The reaction mixture is stirred for 16 hours at 70 C. After cooling to RT the mixture is made basic with 50% aqueous sodium hydroxide solution and saturated sodium hydrogen carbonate solution and extracted with ethyl acetate. The organic phase is washed three times with water, dried on sodium sulphate, filtered and evaporated down i. vac.
