Nonracemic hydroxydihydroisobenzopyranones such as I, analogs of (R)-5-methylmellein, were prepared and tested as inhibitors of monoamine oxidase A (MAO-A). Most of the hydroxydihydrobenzopyranones selectively inhibited MAO-A with IC50 values of 60 nM to 29 μM; I was the most potent and selective analog prepared, with IC50 values of 60 nM for MAO-A and >50 μM for MAO-B.
