To a mixture of 2-amino-5-bromonicotinamide (200 mg, 0.93 mmol), thiophene- 2-carbaldehyde (125 mg, 1.11 mmol) in MeOH (15 mL) was added 4-methylbenzenesulfonic acid (16 mg, 0.09 mmol). The mixture was stirred at 60C overnight. Then the resulting solid was filtered to give the product of 6-bromo-2-(thiophen-2-yl)-2,3-dihydropyrido[2,3- d]pyrimidin-4(lH)-one (180 mg, yield: 63%), which was used for the next step without further purification.
