Researchers who often do experiments know that organic synthesis is a process of preparing more complex target molecules from simple raw materials through one or more chemical reactions. tert-Butyl 4-[(5-bromo-3-nitropyridin-2-yl)amino]piperidine-1-carboxylate. 2,5-Dibromo-3-nitropyridine (0.200 g, 0.71 mmol) was dissolved in DMSO (3 mL) and treated with 4-amino-1-boc-piperidine (0.428 g, 2.14 mmol.) The mixture was heated at 80 C. overnight and then diluted with ethyl acetate (30 mL.) The organic solution was washed with water (3*25 mL), saturated sodium bicarbonate (25 mL) and brine (25 mL). The organic layer was dried over magnesium sulfate, filtered and concentrated under reduced pressure. The crude material was purified by automated flash silica gel chromatography (ISCO Redi-Sep column) eluding with 0-100% ethyl acetate/hexane to yield the title compound. 0.229 g (80%). HPLC (method A): Rt=11.2 min. MS: [M+H-t-butyl]+=344.8.
