In the present study an attempt has been made to formulate Pheniramine maleate a selective H1 receptor antagonist into orodispersible tablet. The tablets were prepared by direct compression method using super disintegrants like croscarmellose sodium, crospovidone, sodium starch glycollate, low hydroxyl pr cellulose and pre gelatinized starch in different ratios. The blend was examined for various pre compression parameters. Tablets were evaluated by measuring hardness, friability, content uniformity, weight variation and drug release pattern. All the tablets met the pharmacopoeial requirements for phys. tests. Almost in all the formulations with increase in concentration of superdisintegrants, the drug release was rapid. Stability studies were also performed. Dissolution studies indicated that the tablets containing crospovidone and croscarmellose sodium showed rapid dissolution compared to other disintegrants releasing almost 100% of the drug in six minutes. Pheniramine maleate (PA), an antihistamine, was determined by Differential Pulse Stripping voltammetry using nano polypyrrole (Ppy) and nano poly (3,4-ethylenedioxythiophene) (PEDOT) modified glassy carbon electrodes. The cyclic voltammetric behavior of pheniramine was studied in aqueous acidic, neutral and alk. conditions. One well-defined oxidation peak was observed in the cyclic voltammograms at all pHs. The influence of pH, scan rate and concentration revealed irreversible electron transfer and the oxidation was diffusion controlled adsorption. The SEM anal. confirmed good accumulation of PA on the electrode surface. A systematic study of influence of various exptl. parameters that affect the stripping voltammetric response was carried out and the maximum peak current conditions were arrived at. Calibration was made under maximum peak current conditions. In this work, a novel TLC chromatog. method coupled with densitometric detection was established for the simultaneous quantification of naphazoline HCl, pheniramine maleate and three of their official impurities, namely; naphazoline impurity B, pheniramine impurities; A & B. Chromatog. separation was carried out on TLC aluminum silica plates F254, as a stationary phase, using methanol: Et acetate: 33.0% ammonia (2.0: 8.0: 1.0, by volume), as a mobile phase. Plates were examined at 260.0 nm and International Council for Harmonisation (ICH) guidelines were followed for method’s validation. Important factors, such as; composition of mobile phase and detection wavelengths were optimized. Linearity was achieved over the ranges of 2.0-50.0μg band-1 for naphazoline, 10.0-110.0μg band-1 for pheniramine, 0.1-10.0μg band-1 for naphazoline impurity B and 2.0-50.0μg band-1 for both pheniramine impurities. The proposed method was assessed in terms of accuracy, precision and robustness where satisfactory results (recovery % ≈ 100% and RSD < 2) were obtained. The method was also applied for the simultaneous determination of naphazoline HCl and pheniramine maleate, in Naphcon-A eye drops, with resp. recoveries of 101.36% and 100.94%. Method greenness was evaluated and compared to the reported HPLC one via environmental, health and safety tool. The mobile phase was 0.1 mol·L-1 ammonium acetate buffer(0.5% triethylamine, adjusted to pH 4.0 with acetic acid)-methanol solution=97:3(V/V), with velocity of 1 mL·min-1 and column temperature of 20°C. The good linearity of pheniramine maleate under chromatog. was found(r>0.999, n=7). The recovery values were between 97% and 103%(n=3). The RSD values were below 2.0%(n=3). The sample was stable in 24 h. The samples reached standardized qualification. RP-HPLC is proved to be a simple, swift, accurate and reliable method and has good reproducibility. Selection of elderly patients (aged ≥60 years) for intensive chemotherapy treatment of acute myeloblastic leukemia (AML) remains challenging. Several cooperative groups such as Acute Leukemia French Association (ALFA), Haematol. Oncol. Clin. Studies Group (HOCSG) and MD Anderson Cancer Center (MDACC) have developed predictive models to select those patients who can benefit from intensive chemotherapy. Our purpose is to validate and compare these three models in a cohort of patients treated in real-life setting. For this, a total of 1724 elderly AML patients and treated with intensive chemotherapy regimens were identified in the PETHEMA registry. Median age was 67.2 years (range, 60-84,9) and median overall survival [OS] 9 mo (95% confidence interval [CI], 8.2-9.7). Taking into account the ALFA group’s model, patients likely to benefit from intensive chemotherapy had longer OS (14 mo, 95% CI 12.3-15.7) than those unlikely to benefit (5 mo, 95% CI 4.1-5.9; p < 0.001). Significant differences in OS were observed between patients with favorable risk (17 mo, 95% CI 13.2-20.7), intermediate risk (11 mo, 95% CI 9.3-12.6) and adverse risk (6 mo, 95% CI 5.1-6.4; p < 0.001) according to the HOCSG model. No significant differences in OS were observed between patients with 0, 1, 2 or ≥3 points according to the MDACC model. However, when patients with ≥1 point were compared with those with 0 points, median OS was significantly longer in the latter [15 mo (95% CI 12.1-17.8) vs 7 (95% CI 5.7-8.5)]. This retrospective study validates predictive models proposed by the ALFA, HOCSG and MDACC groups in this real-life cohort.
