Schiff bases (SB) obtained from S-methyl dithiocarbazate and aromatic aldehydes: salicylaldehyde (H2L1), ovanillin (H2L2), pyridoxal (H2L3) and 2,6-diformyl-4-methylphenol (H3L4), and their corresponding Zn(II)complexes (1-4), are synthesized. All compounds are characterized by elemental analyses, infrared, UV-Vis, nuclear magnetic resonance spectroscopy and mass spectrometry. The structures of H2L2 and [Zn-2(L-1)(2)(H2O) (DMF)] (1a) (DMF = dimethylformamide) are solved by single crystal X-ray diffraction. The SB coordinates the metal center through the Ophenolate, Nimine and Sthiolate atoms. The radical scavenging activity is tested using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, with all ligand precursors showing IC50 values similar to 40 mu M. Cytotoxicity studies with several tumor cell lines (PC-3, MCF-7 and Caco-2) as well as a non-tumoral cell line (NHDF) are reported. Interestingly, 1 has relevant and selective antiproliferative effect against Caco-2 cells (IC50 = 9.1 mu M). Their antimicrobial activity is evaluated in five bacterial strains (Klebsiella pneumoniae, Acinetobacter baumannii, Listeria monocytogenes, Pseudomonas aeruginosa and Staphylococcus aureus) and two yeast strains (Candida albicans and Candida tropicalis) with some compounds showing bacteriostatic and fungicidal activity. The minimal inhibitory concentration (MIC90) of HnL against Mycobacterium tuberculosis is also reported, with H2L2 and H3L4 showing very high activity (MIC90 < 0.6 mu g/mL). The ability of the compounds to bind bovine serum albumin (BSA) and DNA is evaluated for H3L4 and [Zn-2(L-4)(CH3COO)] (4), both showing high binding constants to BSA (ca. 106 M 1) and ability to bind DNA. Overall, the reported compounds show relevant antitumor and antimicrobial properties, our data indicating they may be promising compounds in several fields of medicinal chemistry. Background De novo lipogenesis (DNL) is the primary metabolic pathway synthesizing fatty acids from carbohydrates, protein, or alcohol. Our aim was to examine associations of in vivo levels of selected fatty acids (16:0, 16:1n7, 18:0, 18:1n9) in DNL with incidence of type 2 diabetes (T2D). Methods and findings Seventeen cohorts from 12 countries (7 from Europe, 7 from the United States, 1 from Australia, 1 from Taiwan; baseline years = 1970-1973 to 2006-2010) conducted harmonized individual-level analyses of associations of DNL-related fatty acids with incident T2D. In total, we evaluated 65,225 participants (mean ages = 52.3-75.5 years; % women = 20.4%62.3% in 12 cohorts recruiting both sexes) and 15,383 incident cases of T2D over the 9-year follow-up on average. Cohort-specific association of each of 16:0, 16:1n7, 18:0, and 18:1n9 with incident T2D was estimated, adjusted for demographic factors, socioeconomic characteristics, alcohol, smoking, physical activity, dyslipidemia, hypertension, menopausal status, and adiposity. Cohort-specific associations were meta-analyzed with an inverse-varianceweighted approach. Each of the 4 fatty acids positively related to incident T2D. Relative risks (RRs) per cohort-specific range between midpoints of the top and bottom quintiles of fatty acid concentrations were 1.53 (1.41-1.66; p< 0.001) for 16:0, 1.40 (1.33-1.48; p< 0.001) for 16:1n-7, 1.14 (1.05-1.22; p = 0.001) for 18:0, and 1.16 (1.07-1.25; p< 0.001) for 18:1n9. Heterogeneity was seen across cohorts (I-2 = 51.1%-73.1% for each fatty acid) but not explained by lipid fractions and global geographical regions. Further adjusted for triglycerides (and 16:0 when appropriate) to evaluate associations independent of overall DNL, the associations remained significant for 16:0, 16:1n7, and 18:0 but were attenuated for 18:1n9 (RR = 1.03, 95% confidence interval (CI) = 0.94-1.13). These findings had limitations in potential reverse causation and residual confounding by imprecisely measured or unmeasured factors. Conclusions Concentrations of fatty acids in the DNL were positively associated with T2D incidence. The kinetics and mechanism of condensation of pyridoxal hydrochloride with L-alpha-asparagine, L-alpha- and D-alpha-aspartic acids are analyzed via UV spectroscopy and polarimetry. It is found that L-alpha-asparagine containing alpha-NH2 and gamma-NH2 groups interacts with pyridoxal via the gamma-NH2 group, forming Schiff bases that are resistant to chemical transformations. Rearrangement produces Schiff bases that form the cyclic structure from the amino acid moiety. L-alpha- and D-alpha-aspartic acids interacting with pyridoxal via alpha-NH2 groups create Schiff bases that form quinoid structures after elimination of alpha-hydrogen or CO2. Their subsequent hydrolysis results in pyridoxamine, alpha-ketoacids, and aldehyde acids, respectively. Schemes of the condensation mechanisms of L-alpha-asparagine, L-alpha-, D-alpha-aspartic acids with pyridoxal hydrochloride are proposed.

Background/Aims: Hydrazone and acylhydrazone derivatives, which are produced from aldehyde reacting with hydrazine or acylhydrazine, have been reported to exhibit antitumor activities. However, the angionenic effects of this kind of derivatives haven’t been elucidated. Here, we synthesized 12 pyridoxal hydrazone and acylhydrazone compounds and investigated their antiangiogenic effects and the underlying mechanisms. Method: 3-(4,5-Dimethylthiazol-2-yl)-2, 5-dipheyltetrazolium bromide assay was used to screen the inhibitory effects of the synthesized compounds on endothelial cells (ECs) proliferation. The compound with best inhibitory effect was further evaluated with wound-healing assay and tube formation assay. Calcein-Am assay was carried out to determine the content of intracellular labile iron pool (LIP). Intracellular reduced glutathione (GSH) was determined by spectrophotometry. Flow cytometry was used to determine cell cycle and apoptosis. Results: Compound 10 (3-hydroxy-5-[hydroxymethyl]-2-methyl-pyridine-4-carbaldehyde-2-naphthalen-1-acetyl hydrazone) showed the best inhibitory effect on human umbilical vascular ECs proliferation, with IC50 value of 25.4 mu mol/L. It not only inhibited wound-healing and tube formation of ECs, but also decreased the content of intracellular LIP and GSH. Furthermore, it arrested ECs cycle at S phase and induced cell apoptosis. Conclusions: Compound 10 exhibits antiangiogenic effects by reducing the content of intracellular LIP and GSH, and subsequently arresting cell cycle and inducing cell apoptosis.

A chemical sensor composed of pyridoxal hydrochloride schiff base based on diarylethene (1O) was synthesized. Its photochemical properties and selectivity to ions were further studied. The chemosensor could detect cyanide effectively and is almost undisturbed by other ions. When titrating CN-, the reaction aroused a distinct change in the absorption spectrum with the color change from transparent to yellow, and the fluorescence intensity centered at 562 nm was increased 68 folds. It also exhibited a good fluorescence sensing of Mg(2+ )with high selectivity and sensitivity. Upon addition of Mg2+, its emission intensity enhanced 110 folds, with the color change from dark to bright blue. Its good spectral response could be applied to molecular logic circuit. Moreover, the chemosensor could be made into test paper strips for the qualitative and quantitative detection of CN- and Mg2+.

Mixed ligand nickel(II) and ruthenium(II) complexes were synthesized from pyridoxal-N-allyl-thiosemicarbazone hydrochloride and triphenylphosphine. The structures of the complexes have been characterized by elemental analysis, IR, H-1 and P-31 NMR, conductivity, magnetic moment measurements and single-crystal X-ray diffraction technique. Based on X-ray crystallographic studies, a square-planar structure has been proposed for the Ni(II) complex, in which the thiosemicarbazone ligand acts as dianionic tridentate ONS ligand. In the case of the Ru(II) complex, the thiosemicarbazone is coordinated to metal atom as a monoanionic bidentate NS donor ligand in an octahedral geometry. Antioxidant activities of the ligand and its metal complexes were calculated as their trolox equivalent antioxidant capacities (TEAC) by CUPRAC method and DPPH assay. Both the ligand and its metal complexes were found to be antioxidant and are much more antioxidant at least 2.1 times than trolox, even, ligand is 3.5 times greater than that of trolox according to CUPRAC. A linear correlation (correlation coefficient R-2 = 0.9997) appeared between the obtained TEAC values by the two antioxidant assays.

A new unsymmetrical N2O2-tetradentate Schiff-base complex of zinc(II) was synthesized by the template reaction of pyridoxal-S-methylthiosemicarbazone and 2-hydroxy-4-methoxy-benzaldehyde as starting compounds. S-methylthiosemicarbazone (1) and zinc(II) complex [Zn(L)CH3OH] ( 2) were characterized by elemental analysis, FT-IR, UV-visible, H-1, and C-13 NMR spectra. The molecular structure of the complex (2) was determined by single crystal X-ray diffraction technique. The structure consists of a distorted square-pyramidal geometry around the central metal, Zn(II). Quantum chemical calculations were carried out using density functional theory DFT/B3LYP, 6-31G (d), and LanL2DZ basis sets for theoretical characterization of the compounds. The experimental and theoretical data were compared comprehensively. The potential energy distribution (PED) analysis was performed for the assignment of vibration frequencies. In order to support in vitro studies, molecular docking studies have been carried out so that the title compound can be an inhibitor of Epidermal Growth Factor Receptor (1 m17), and the relationship between calculated HOMO energies and docking studies has been examined. In addition, the total antioxidant capacity (as TEAC value) and free radical scavenging activity of the compounds were determined by Cupric Reducing Antioxidant Capacity (CUPRAC) and 1,1-diphenyl-2-picryl hydrazyl (DPPH) methods, respectively. Small-scale culture of animal cells in suspension is of importance for many applications. At a small-scale, fed-batch is achieved either by manual bolus feeding or the use of liquid handling robots. In this study, we report an alternate application of a hydrogel for in situ continuous delivery of a nutrient feed comprising 18 amino acids, vitamins, antioxidants, and trace elements. We show that amino acid release is sustained for at least seven days. Importantly, release rates of individual amino acids can be independently modulated by changing their loading. We demonstrate the application of this hydrogel for complete in situ feeding of nutrients to a suspension adapted CHO cell line expressing IgG leading to 2.7-fold and 4-fold improvement in integral viable cell density (IVCD) and volumetric productivity respectively. This is similar to improvements obtained by bolus liquid feeding. Further, supplying glucose from the same hydrogel to eliminate manual feeding led to a 1.8-fold increase in IVCD accompanied by a 3-fold increase in volumetric productivity as compared to batch culture. In summary, this study provides a proof-of-concept that hydrogels can enable completely closed in situ feeding for mammalian cell culture requiring no external intervention. Such continuous in situ delivery can potentially enable closed culture systems maintaining nutrients at low levels mimicking physiological concentrations.

Three hydroxymethyl bridged Cu(ii) complexes of a pyridoxal Schiff base ligand 4-((E)-(2-(pyridin-2-yl)ethylimino)methyl)-5-(hydroxymethyl)-2-methylpyridin-3-ol (LH) have been synthesized and characterized on the basis of spectroscopic, elctrochemical and structural properties. The X-ray crystal structures of the complexes reveal dual denticity of the ligand, bidenticity in the absence of a co-ligand as in complex1, and tridenticity in the presence of a co-ligand such as SCN-/N(CN)(2)(-)as in complexes2and3. The complexes, though binuclear in the solid state, exist as a monomeric unit in solution due to the exceptionally long axial Cu-O-hydroxymethyl(2.4-2.5 angstrom) bond. All three complexes show efficient catalytic activities towards the aerial oxidation of 3,5-ditertiarybutylcatechol (DTBCH2) withk(cat)values of 5.38 x 10(4)h(-1), 1.18 x 10(5)h(-1)and 1.06 x 10(5)h(-1)in methanol. Complexes1and2also act as a selective sulphide ion sensor withK(b)values of 6.6 x 10(3)M(-1)and 8.1 x 10(3)M(-1), respectively, while their respective L.O.D. values are 3.4 mu M and 3.2 mu M.
